The Interaction of Carcinogenic Hydrocarbons with Tissue Constituents

I. Methods* ,{dagger}

Walter G. Wiest{ddagger} and Charles Heidelberger

( McArdle Memorial Laboratory, The Medical School, University of Wisconsin, Madison 6, Wis.)

Procedures have been developed for the homogenization of mouse skin and submaxillary gland. This has made possible the separation of submaxillary gland by differential centrifugation into its cellular components and also the determination of the number of cells, the nucleic acid content, and the quantity of protein nitrogen per unit of tissue. Methods have been described for the administration of labeled carcinogenic hydrocarbons to these tissues and for the determination of radioactivity in the isolated fractions and proteins.

* An abstract of this report appeared in Cancer Research, 12: 308, 1952.

{dagger} This work was supported by a grant from the Wisconsin Section of the American Cancer Society, by a grant-in-aid from the National Cancer Institute, and by the Pearl and W. M. Quandt Memorial Fund.

{ddagger} Present address: Department of Biochemistry, University of Utah Medical School, Salt Lake City, Utah.

Received 10/20/52.
