Quantitative immunological studies have been made with carcinogen-protein conjugates containing widely different numbers of 4-dimethylaminostilbenyl- and 2'-methyl-4-dimethylaminostilbenyl-4'-carbamido groups attached to bovine and horse serum albumins. The serological reactivity of the protein component was found to decrease progressively with increased substitution of carcinogen. The introduction of four prosthetic groups conferred only slight haptenic activity on the immunizing antigens, and 1314 groups represented the lower limit for demonstration of haptenic activity in the test antigens. Conjugates containing 1337 carcinogen groups retained satisfactory immunizing capacity and displayed high prosthetic group activity as immunizing agents. A pronounced drop in immunizing capacity was caused by the introduction of 5562 stilbenyl groups into the proteins. The influence of light in decreasing the intensity of absorption and in promoting trans-cis shifts in the stilbenyl prosthetic groups has been taken into consideration in the interpretation of the immunological results.
