Studies in 2-Acetylaminofluorene Carcinogenesis

I. The Intracellular Distribution of Nucleic Acids and Protein in Rat Liver*

1. The course of AAF carcinogenesis (liver) involved a progressive increase in the percentage of cell protein and pentosenucleic acid in the nuclear fraction and a concomitant, but more drastic, reduction of the fraction of these constituents associated with the mitochondria.
2. During the preneoplastic period, no increases were noted in DNA, but an increase of 50 to 100 per cent, corresponding to the increased cellularity, was observed in the tumor.
3. Alterations in the chemical composition of the nucleus and mitochondria were observed during carcinogenesis. These resulted in an increase in the protein/DNA and the PNA/DNA ratios of the nucleus.
4. The alterations in cellular morphology and composition have been discussed from the point of view of a self-accelerating disturbance in the cytoplasmic-nuclear relations leading to the establishment of a new balance in the neoplastic state.
