The Effects of A-Methopterin on the in Vitro Incorporation of P32O4 into Normal and Leukemic Mouse Tissues*

( Department of Biochemistry and Nutrition, University of Southern California, Los Angeles, Calif., the Department of Biological Chemistry, University of Illinois College of Medicine, Chicago, Ill., and the National Cancer Institute, Bethesda, Md.)

The in vitro uptake of P32O4 into the phosphoprotein, phospholipid, and pentosenucleic acid nucleotides of normal DBA mouse liver and transplantable lymphocytic leukemias sensitive or resistant to folic acid antagonists was studied. The action of a folic acid antagonist, A-methopterin, on this uptake showed no significant effect on P32O4 incorporation into any of the constituents studied. This suggests that direct interference with nucleic acid phosphorus metabolism is not involved in the antileukemic action of the folic acid antagonists.

* This investigation was supported by a research grant from the National Cancer Institute of the National Institutes of Health, Public Health Service.

{dagger} Predoctoral research fellow of the National Cancer Institute of the National Institutes of Health, Public Health Service.
