It was found that progressive loading of the reticulo-endothelium in mice by intravenous injections of Thorotrast was followed by significant changes in the intraperitoneal growth rate of the Ehrlich ascites tumor. With small dosages, the cell multiplication rate became depressed to a degree which is probably significant. With larger dosages the tumor grew faster than in the control animals. One high point in the multiplication rate was reached at the 3-dose level; the rate then fell somewhat through the 5-dose level. Beyond this point the increase ratio again rose significantly with increasing dosage of the colloid. An explanation of the above findings is proposed on the basis of reticulo-endothelial inhibition of tumor growth, the control mechanism being modified by increasing demand made on phagocytic activity.
* The investigation reported in this paper was sponsored in part by a grant-in-aid from the U.S.P.H.S. The Thorotrast used was a gift of the Heyden Chemical Corp. The statistical treatment of data was carried out under the guidance of Dr. Huldah Bancroft, Professor of Biostatistics, Tulane University School of Medicine.
