A method for the continuous cultivation of Novikoff rat hepatoma cells In vitro is described. Multiplication rates of the cells were measured by cell counts, with different nutrients. Optimal growth occurred in a mixture containing 0.5 per cent lactalbumin hydrolysate and 5 per cent calf serum, in Earle's saline. Of several viruses tested, none caused detectable cytopathogenic effect on the cells. In calf serum the cells grew rapidly, generally in a rounded condition, whereas in normal rat serum the cells formed a closely packed sheet.
After In vitro cultivation for 8 weeks the cells were still able to cause typical tumor production after intraperitoneal inoculation into an adult rat.
