Clusters of morphologically transformed cells (foci) resulting from infection of secondary cultures of chick embryo cells by Rous sarcoma virus (RSV) were compared with fibroblast-like cells outside the focal areas with the use of several cytochemical procedures. Transformed cells yielded greater precipitation of the dinitroformazan in a test involving the lactic acid dehydrogenase, NBT-reductase complex. Intracytoplasmic fluorescent material was observed in these cells following treatment with fluorescein-labeled anti-RSV serum. Differences were observed in the utilization of labeled precursors for nucleic acid synthesis following short exposures to uridine-H3 and thymidine-H3.
