The possibility of obtaining differential protection of normal and malignant tissues against radiation in tumor-bearing mice by pretreatment with 2-mercaptoethylguanidine has been investigated. Although neither the mammary tumor nor the leukemia studied appeared to be protected against local x-ray exposure at sites of transplantation, it was not possible to greatly improve the therapeutic index of whole-body x-radiation by this means. In the case of the mammary tumor the whole-body x-radiation dose that could be administered was still below the level needed to restrain tumor growth. Results obtained in the treatment of the more radiosensitive leukemia suggested that different routes of administration resulting in a more favorable metabolism and distribution of protective compound may improve these results.
