A new anti-leukemic agent, 1,3-bis (2-chloroethyl)-1-nitrosourea (NSC-409962), has been found to be a potent inhibitor of nonproliferating microorganisms. Comparative studies with 1-methyl-1-nitrosourea (NSC-23909) and 1-methyl-3-nitro-1-nitrosoguanidine (NSC-9369) strongly suggest a similarity in mechanism of action. A strain of Escherichia coli resistant to NSC-409962 is cross-resistant to mitomycin C, porfiromycin, several alkylating agents, azaserine, 6-diazo-5-oxo-L-norleucine, a terephthalanilide (NSC-60339), ionizing and ultraviolet irradiation, but not to 6-mercaptopurine or amethopterin. Strains of E. coli resistant to mitomycin C, porfiromycin, nitrogen mustard, or azaserine were cross-resistant to NSC-409962.
