The effect of actinomycin on radioautographic RNA and protein labeling and ultrastructure was studied in the liver of rats after partial hepatectomy. Actinomycin almost eliminated nucleolar RNA labeling and strongly inhibited cytoplasmic RNA labeling. The protein labeling was reduced only over nucleoli. The hepatectomized, untreated animals showed large nucleoli and hypertrophic endoplasmic reticulum. The hepatectomized, actinomycin-treated animals showed small, collapsed nucleoli and an endoplasmic reticulum similar to that of nonoperated animals.
