3-Methylcholanthrene (3-MC)2 was incubated with syngeneic mouse liver, spleen, kidney, and muscle for 16 hr at 37°C to which CFA was added 1:1. Eight groups of C3H Bittner virusfree male mice were used in the study. Group A was pretreated with this material. Group B was pretreated with a preparation which was refrigerated 16 hr at 4°C, to slow the rate of chemical reactions, instead of being incubated for 16 hr. Group C received no pretreatment. Group D was pretreated with 3-MC-incubated nuclei from spleen, liver muscle, and CFA. Three weeks later the 4 groups of mice, A, B, C, and D, were implanted with a paraffin pellet containing 800 µg of 3-MC. Group E served as spontaneous tumor control mice which received no pretreatment and no 3-MC.
Groups F, G, and H were pretreated with the same material as A, B, C, and D, but with saline substituted for CFA.
The mean tumor incidence was significantly lower in Group A mice pretreated with the 3-MC tissue and CFA when compared to Group C which received 3-MC alone in a carcinogenic dose. Group B mice pretreated with the CFA-tissue-3-MC preparation refrigerated instead of incubated responded in the same way as Group C which were not pretreated. Group D mice pretreated with nuclei, instead of whole tissue, responded in the same way as Group A. The presence of CFA in the pretreatment preparation appeared to enhance host resistance to cancer.
