The biosynthesis of DNA and the activities of enzymes involved in this pathway were investigated in the surviving rat kidney after unilateral nephrectomy. It was shown by radioautographic technics that DNA synthesis is markedly enhanced by 24 hours and reaches a peak at 48 to 72 hours postoperatively. Deoxythymidine (thymidine; thymine deoxyribonucleoside) kinase, deoxythymidine monophosphate kinase, and 2'-deoxycytidine-5'-monophosphate (dCMP) deaminase also increased in activity, reaching a maximum at 24 to 48 hours postoperatively, whereas 2'-deoxythymidine-5'-monophosphate phosphatase and dCMP kinase did not show any consistent change. These changes were observed just prior to the burst in mitotic activity as measured by radioautographic technics.
