3,4-Benzpyrene, 7,12-dimethylbenz(a)anthracene, and 1,2-benzpyrene were topically applied to mouse skin in order to study their effects upon RNA synthesis in the latter. Both of the known carcinogenic substances [3,4-benzpyrene and 7,12-dimethylbenz(a)anthracene] caused a preliminary decrease in the rate of RNA synthesis, followed by an abrupt increase well above the normal rate; however, the 2 curves differ in shape, the preliminary decrease occurring more rapidly in the case of the 3,4-benzpyrene. The early inhibitory effect was not observed for the noncarcinogenic 1,2-benzpyrene, the only occurrence being an increase in the rate of RNA synthesis. The electrophoretic study of skin RNA showed no differences between the RNA patterns for treated and for normal skin.
