Transplantable Rat Mammary Tumors Induced by 5-Nitro-2-furaldehyde Semicarbazone and by Formic Acid 2-[4-(5-Nitro-2-furyl)-2-thiazolyl]hydrazide1

Mammary fibroadenomas induced by 5-nitro-2-furaldehyde semicarbazone and mammary adenocarcinomas induced by formic acid 2-[4-(5-nitro-2-furyl)-2-thiazolyl]-hydrazide in Holtzman female rats were subcutaneously transplanted into male and female newborn Holtzman rats to determine the biological activity of these tumors. Fibroadenomas induced by 5-nitro-2-furaldehyde semicarbazone were transplantable and showed increased anaplasia, pleomorphism, and mitotic activity. Increased malignancy was evidenced by the development of an adenocarcinoma in a rat inoculated with a fibroadenoma.

Since the 5-nitro-2-furaldehyde semicarbazone used to induce the tumors utilized in the transplantations contained 3% of 5-nitro-2-furaldehyde azine as an impurity, pure 5-nitro-2-furaldehyde semicarbazone was retested. A high incidence of mammary tumors (22/29) was observed. 2-Furaldehyde semicarbazone was also tested and only 6 of 30 rats developed mammary fibroadenomas. The incidence of these tumors was not statistically greater (p > 0.14) than that observed in the control rats (2/29) that developed histologically identical mammary fibroadenomas. These data suggest that the nitro group of 5-nitro-2-furaldehyde semicarbazone is of primary importance in the genesis of these tumors.
