Studies were conducted on mitochondria from B16 amelanotic melanoma cells. The isolated mitochondria showed no contamination of premelanosomes, and the structures were well preserved. Intermediates of the tricarboxylic acid cycle, pyruvate,
-glycerophosphate, and tetramethyl-p-phenylenediamine, were oxidized by these mitochondria with a good coupling of O2 uptake and ADP phosphorylation. The ADP:O ratios were normal for all substrates tested except
-ketoglutarate, which showed an ADP:O ratio of 3 or lower. Spectrophotometric measurements at liquid nitrogen temperature showed that the a, b, and c type cytochromes content of these mitochondria were similar to those of other mammalian mitochondria. It was concluded that the qualitative characters of mitochondria from amelanotic melanoma were essentially similar to those of normal, nonneoplastic cell mitochondria.
