Undermethylated Escherichia coli tRNAPhe has reduced activity in aminoacylation when measured by differences in initial velocities; however, the Km remains unchanged. The conformation of undermethylated tRNAPhe, measured by optical methods, is different from its normal counterpart. The ability of undermethylated tRNAPhe to support poly U-stimulated polyphenylalanine synthesis is 3-fold less than the normal molecule.
