Interferon production resulting from Newcastle disease virus stimulation of cultured leukocytes was measured in cells derived from 21 consecutive, untreated, newly diagnosed patients with Hodgkin's disease and in a group of control subjects. In addition, skin tests and phytohemagglutinin-induced lymphocyte blastogenesis were assessed. It was found that interferon production was normal in cells from Stage I, II, and III patients but was impaired in Stage IV patients. The interferon titer produced by the cultured leukocytes was found to be related to the absolute number of lymphocytes as well as glass-adherent cells in the cultures. Cultures prepared from the Hodgkin's patients had a decreased mean mononuclear cell number, but this alone did not account for the impaired responses observed in Stage IV patients. There was no correlation between the phytohemagglutinin response as measured by tritiated thymidine uptake and the interferon produced in response to virus. Likewise, no correlation was found between lymphocyte interferon production and the presence of anergy. Impaired interferon production was observed to return to normal in those patients achieving remission.
