Effect of Vincristine on Sister Chromatid Exchanges of Normal Human Lymphocytes1

Claude Stoll2, Digamber S. Borgaonkar and Jean-Marc Levy

Divison of Medical Genetics, Department of Medicine, The Johns Hopkins University School of Medicine, Baltimore, Maryland 21205 [C. S., D. S. B], and Service de Pediatrie IV, C.H.U., Strasbourg, France [C. S., J-M. L.]

In vitro analysis of the mitosis from 3-day lymphocyte cultures from two normal males and two normal females treated with bromodeoxyuridine and stained with acridine orange after addition of vincristine during different times demonstrated that vincristine inhibits the progression of the second division. When the second division progressed, the number of sister chromatid exchanges was significantly decreased. Vincristine could act by inhibiting the copying of DNA.

A dosage effect is demonstrated when vincristine is added to the cultures during the first 24 hr.

1 These studies were supported in part by USPHS Grant GM 19489 Genetics Center Grant.

2 Recipient of Eli Lilly postdoctoral fellowship.

Received 9/ 5/75. Accepted 4/14/76.
