Fig. 6. Antitumor effect of 8H9(Fv)-PE38 in severe combined immunodeficient mice. Groups of animals received injection with 2 x 106 MCF-7 cells (A-D) or OHS-M1 cells (E and F) on day 0. On day 4, tumors reached a size of 50 mm3. Animals were treated i.v. on days 4, 6, and 8 with 0.075 ({triangleup}) and 0.15 mg/kg ({blacktriangleup}) 8H9(scFv)-PE38 in Dulbecco’s modified PBS containing 0.2% HSA or 0.075 ({circ}) and 0.15 mg/kg ({bullet}) 8H9(dsFv)-PE38 in Dulbecco’s modified PBS (0.2% HSA). Control groups received diluent alone ({blacksquare}) or M1(dsFv)-PE38 ({square}), which is an immunotoxin against CD25. No deaths were observed at these doses. Comparison of tumor size (*) between {blacksquare} and {triangleup}, {blacktriangleup}, or {circ} gives P < 0.05. Data are expressed as the mean ± SD (n = 5 or 10).
